Mozilla Labs has released the first beta of Thunderbird 5 (formerly Thunderbird 3.3) on June 2, 2011. To bypass extension compatibility checking in Thunderbird 5 Beta 1:
- Tools Menu > Options…
- Advanced Tab > In the Advanced Configuration section near the bottom click the Config Editor… button
- Click the I’ll be careful, I promise! button
- Right-click in the list and select New > Boolean
- Enter extensions.checkCompatibility.5.0b and set to FALSE
- Close the about:config Windows and click OK on the Options window
- Close and restart Thunderbird and previously disabled add-ons should automatically be re-enabled. If not go to Tools Menu > Add-ons
The latest version of Thunderbird Beta has the following changes:
- New Addons Manager and extension management API (user interface will be changed before final release)
- Tabs can now be reordered and dragged to different windows
- Revised account creation wizard, offering improved set-up
- Attachment sizes now displayed along with attachments

A quick reminder Thunderbird 4 has been skipped in an effort to keep the current Thunderbird numbering in-line with that of Firefox. Not sure yet when the final Thunderbird 5 will be released, but will likely be sometime around June 21st-28th.
